Inoculation of airborne conidia of Penicillium chrysogenum on the surface of a solid medium

International audience; To reproduce a fungal contamination of food products by airborne conidia, a method to inoculate a few number (in the range 1-9) of conidia on the surface of agar media was developed. This technique would allow to determine accurately the time to detection of fungal colonies, then the mould free shelf-life of food products by using dry conidia. The method was based on dry-harvesting the conidia in the lid by gently taping the bottom of the dishes where sporulating mycelium was grown, retaining the conidia on glass beads, and, aseptically transferring the beads to successive Petri dishes to "dilute" the samples. Alginate/silica composite hydrogel as a potential morphogenetically active scaffold for three-dimensional tissue engineering

Pursuing our aim to develop a biomimetic synthetic scaffold suitable for tissue engineering, we embedded bone cells, osteoblast-related SaOS-2 cells and osteoclast-like RAW 264.7 cells, into beads, formed of a Na-alginate-based or a silica-containing Na-alginate-based hydrogel matrix. The beads were incubated either separately (only one cell line in a culture dish) or co-incubated (SaOS-2-containing beads and RAW 264.7 beads). The alginate and alginate/silica hydrogel matrices were found not to impair the viability of the encapsulated cells. The efficacy of ethylene-vinyl alcohol copolymer films (EVOH) incorporating the essential oil components cinnamaldehyde (CINHO), citral (CIT), isoeugenol (IEG), or linalool (LIN) to control growth rate (GR) and production of T-2 and HT-2 toxins by Fusarium sporotrichioides cultured on oat grains under different temperature (28, 20, and 15 °C) and water activity (aw) (0.99 and 0.96) regimes was assayed. Lactate-induced inhibition of tumor cell proliferation.

Abstract Culture medium that was recovered from tumor cell or fibroblast cultures during the plateau phase, and that was replenished by addition of glucose, glutamine, and serum and readjustment of pH had a distinct growth-inhibiting effect on monolayer cell cultures. The effect, which was not specific for a given cell strain, may be partially responsible for the "density inhibition" commonly observed in malignant cells grown in monolayer cultures. The selection of serum-independent PC12 cells for a more-reliable manganese cytotoxicity test.

A major issue concerning the protocols of heavy metal cytotoxicity tests with PC12 cells was the hypothesis that serum in the culture medium might sequester the metal, thus altering the results obtained. However, serum withdrawal impairs the viability of PC12 cells themselves, thus impeding cytotoxicity testing in the absence of serum. In this study, we repeatedly selected undifferentiated, totally non-adherent PC12 cells in Petri dishes. Cell-cell recognition system in gorgonians: description of the basic mechanism

The dissociation of the gorgonian Eunicella cavolinii (Koch) into single cells was successfully accomplished. These cells readily formed aggregates of a size of 2 100 μm during incubation in roller tubes; no aggregate formation was observed in non-rotating Petri dishes. The formation of aggregates was not influenced by Ca++, urea or trypsin; it was also independent of temperature (4° to 30°C) and pH (5.5–9.0). We investigated cues that mediate the aggregation behavior of immature pentatomid bugs by using nymphs of six different pentatomid bug species (Nezara viridula, Acrosternum hilare, Chlorochroa ligata, Chlorochroa sayi, Thyanta pallidovirens, and Euschistus conspersus). When first instars of any two species were put together in a Petri dish, they readily formed heterospecific aggregations similar to their natural conspecific aggregations.
